NA-931 (Bioglutide) is a first-in-class, orally administered, small-molecule quadruple receptor agonist targeting GLP-1, GIP, glucagon, and IGF-1 receptors, designed for the treatment of obesity and metabolic disorders.  Based on company-reported Phase 2 clinical trial data, it demonstrates several potential health benefits:

  • Significant Weight Loss: In a 13-week, double-blind, placebo-controlled Phase 2 trial, NA-931 produced dose-dependent body weight reductions of up to 13.8% at the 150 mg daily dose, with 72% of treated subjects achieving at least 12% weight loss—compared to just 1.9% in the placebo group.
  • Preservation of Lean Muscle Mass: Unlike many weight-loss therapies that cause muscle loss, NA-931 showed no muscle loss in clinical trials, attributed to its IGF-1 receptor modulation, which supports anabolic signaling and lean mass maintenance.
  • Improved Glycemic Control: The drug enhances insulin secretion and glucose regulation through GLP-1 and GIP agonism, with potential benefits for individuals with type 2 diabetes and insulin resistance.
  • Increased Energy Expenditure: Glucagon receptor activation is expected to boost metabolic rate and fat oxidation, helping counteract metabolic adaptation during weight loss.
  • Favorable Safety and Tolerability: Reported adverse events were mild or insignificant, with low rates of nausea (7.3%), vomiting (7.3%), and diarrhea (6.3%).  No serious adverse events were observed, and gastrointestinal side effects were comparable to placebo.
  • Convenience of Oral Administration: As a once-daily oral pill, it offers a non-injectable alternative to current GLP-1-based therapies, potentially improving adherence.
  • Potential Benefits for Liver Health: Preclinical data suggest reductions in liver fat and triglycerides, indicating possible benefits for non-alcoholic steatohepatitis (NASH) and metabolic dysfunction-associated steatotic liver disease (MASLD).

Note: While these results are promising, NA-931 has not yet been approved by the FDA or EMA. All current data come from company-issued press releases and conference presentations, with no peer-reviewed publication of full trial results to date.  The IGF-1 receptor agonism claim remains unverified in independent scientific literature.  Clinical development is progressing toward Phase 3 trials.
